[Two Guys] decided that they have spent too much time in paradise and decide to take a vacation to Japan. Through the entirety of the film, there is no major conflict, there is no primary antagonist, there is no massive plot that involves saving the world or some other thing like it. It is a simple, but comic, slice of life story, their small daily adventures while exploring a modern Japanese culture, such as shopping, going to amusement parks, saunas, and pools, with it all ultimately culminating in a celebration of the Christmas holiday and a ushering in of the new year. Saint Young Men, a buddy/room-mate comedy anime about Buddha and Jesus that is perfect for the online GIF culture.

Aw, I think that the Buddha and Jesus ought to be able to spend a day together at Tokyo Disneyland. Actually, I want to spend a day with those two guys at Tokyo Disneyland ("USALand" in the movie).

rebent, a manga fan/translator wrote that Buddha's t-shirt reads "Siddhartha" (Buddha's proper name) and Jesus' reads "amen." I'm not sure if their shirts are constant.

Do you think they know Saint Seiya or Saint Tail?
Jesus is pretty much an otaku, so probably. Buddha got so emotioned by Osamu Tezuka's Buddha when he had to stay overnight at a manga kissa that he's even begun drawing a manga series of his own ("Get illuminated, Ananda!"). He got good at drawing in the past by making sand mandalas.
One funny gag in the comic is when Jesus and Buddha go to Akihabara because it's a place of pilgrimage for Jesus and Buddha needs a rice cooker. At one moment, Jesus picks up a Sailormoon-esque magic wand at a shop and gets a transformation into fluffy Renaissance-style robes. It's that kind of series.
Re: shirts, the manga has them change their shirts every chapter, because Buddha has silkscreening as a hobby and does them all. Eventually, when they're trying to lose weight by jogging around the neighborhood, Jesus dries his sweat with one and it turns into a talking Veronica t-shirt (like in Dokonjou Gaeru).

They're not. The guy at the convenience store ends up keeping a observation log for the shirts.

I am really digging this. I clicked the link to the manga first, and I think when my brain tried to adapt to the manga reading pattern of words are still read left-to-right, but the cells and conversation bubbles are read in a right-to-left order, it short-circuited my brain and I totally missed the link to the actual movie in the main post, but instead ended up finding two half-hour OVA videos I mistook for TV episodes (links to 1 and 2).
They both are very good, and make sure you check out Buddha's dream/nightmare scene that happens in the middle of episode 2 - it's fantastic.
